Is there any way to set up a symbol library in Illustrator and be able to use it in ArcGIS (or vice-versa?). Preferably in a vector format and preferably not through a font

If you don't have too many to do. You can save the symbols you want to use as EMF files out of Illustrator and then import them into ArcMap using the style manager.
From ArcMap select Tools>>Styles>>Style Manager
From here you can add your EMF files into existing style or create your own new style.
After you figure out where you want your symbol to go. You can select that directory on the left and then right click on the white space on the right.
When you right click selct New>>Marker Symbol
In the next dialog under properties set the type to Picture Marker Symbol. This will bring up a dialog that will allow you to navigate to your EMF file.
Once you get all of this set up it actually works quite well. Although if you have hundreds of symbols to do there has to be a better way.
To get symbols from ArcMap to Illustrator I usually just place the symbol somewhere on the ArcMap document using the drawing tools. Then I copy and paste the symbol to Illustrator and add it to Illustrator's symbol pallete from here...
Hope this helps.
